It would be better to use them after every 10th boss, so you get enough time until your next boss fight.Use your first Ascension when you have reached zone 139 that’s the max cap you can hit at your first playthrough.At zone139 farm and level up as much heroes as you can that your total hero level count exceeds 12k to get enough HS.For active play, YOU MUST GET THESE ANCIENTS. Juggernaut (Additive, stacking, DPS bonus for constant clicking), Fragsworth (+20% Click Damage), Bhaal (+15% Critical Damage) and Vaagur (-5% skill cooldown). It doesn’t matter in what order you get these, although it would be preferable to get Juggernaut and Vaagur as your first Ancients.Use your skills in this order to make the most out of these:- 8th “Energize” (Doubles the effect of next skill you use)<7th "Super Clicks" (200% click damage combined with your 8th skill doubles it)<1st "Clickstorm" (I don't find this skill that useful, it is basically your autoclicker enabled for 30seconds only)<2nd "Powersurge" (If you have Jaggu ancient stacked upto 200k combo, with this skill it will increase your overall 100% more DPS which makes it quite useful)<3rd "Lucky Strikes" (If you have unlocked all the heroes till Forstleaf, you should have default 9% critical chance and combining it with "Bhaal Ancient" will do significant amount of damage making it the best skill out of all)<4th "Metal Detector" (Like I said, use your skills only on Boss fights. When you defeat a boss while this skill is active you will get huge amount of gold)<5th "Golden Clicks" (More effective on Boss fights)<6th "Dark Ritual" (2nd best skill, it increase your total DPS by 1.05, but has 8hours of cooldown. If you have picked "Vaagur ancient", he is your new Best friend)<9th "Reload" (Make sure "Dark ritual is the last skill you used before picking this skill. Although, a lot of people prefer it to combined with "Lucky Strikes")Keep farming that boss, unless your cooldown begins.
