How to unlock Lost character? Alright buddy, this guide will help you unlock lost. These steps are pretty straightforward so you should have no problem with them.

IMPORTANT !

If you own Afterbirth the Rebirth method doesn’t work

You need these things to be unlocked before starting.
Sheol (Defeat Mom’s heart 10 times)
Magdalene (Have seven or more heart containers at once in a run)
Judas (Beat Satan once)
Azazel (Make three devil deals in one run)

Unlocking The Lost in Rebirth

1. Die to a Mulliboom in the Basement or Cellar as Isaac. ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎
‏‏‎Mulliboom is in the room under the spawn room. Make sure to have have half heart when you run in.

2. Die to your own bombs in The Caves or Catacombs as Magdalene. ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ Seed: FHRD S1N1 (normal)
‏‏‎‏‏Bomb deals 1 damage.

3. Die to Mom as Judas. ‏‏‎Make sure moms kills you. Avoid getting killed by tiny enemies, if you get killed by random enemy this will reset your progress.
Seed: D0VC 4VDS (normal) Just nice run with judas.

4. Die to the 2nd stage of the Satan boss fight as Azazel. ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎ ‏ ‏‏‎ ‏‏‎ ‏‏‎ ‏‏‎You Need to die in this phase

Unlocking The Lost in Afterbirth

The Lost is unlocked by dying in a Sacrifice Room while holding Missing Poster. To unlock Missing poster you have to defeat boss The Lamb as Isaac.
